Biospace Lab PhotonIMAGER 小动物活体光学系统有创新的成像方式,4D Real-Time 实时成像能力是它的独家优势技术。传统的小动物活体光学成像,输出的是单张照片。将实验小动物放入成像舱后,根据标准曲线推测出信号高峰时间,在这个节点上拍摄照片,输出拍摄时刻小动物的光学信号图像。然而,传统的成像方式无法应对小动物的个体差异,也无法展示小动物体内信号变化的信息,然而Biospace Lab PhotonIMAGER 的 4D Real-Time 实时成像功能则是输出一段视频,展示在整个拍摄过程中,小动物体内实时的信号变化,包含了强度信息、位置信息和时间信息,这得益于Biospace Lab的核心技术:iCCD技术。iCCD技术使得Biospace Lab有着比市面上同类产品更高的灵敏度(37 ph/s/sr/cm2)、更快的时间分辨率(66 fps)以及更高的信噪比,完全无需binning就可以获得非常清晰的图像。

也因此,Biospace Lab PhotonIMAGER可以做到5D小动物清醒成像。小动物在成像暗舱中自由活动,消除了麻醉带来的影响,输出更真实的小动物活体光学数据,拓展了在神经学、代谢学和动物行为学方面的应用。

另外Biospace Lab还具有创新的模块化成像方式,除了上述的5D清醒成像模块,还有3D成像模块、X光成像模块及大动物成像模块,可以根据实际需求对设备进行定制、随时随地的原地升级、任意切换模块,提供了更便捷的可选方案。


1. 小动物活体光学成像系统研究应用领域

1、癌症与抗癌药物研究

直接快速地测量各种癌症模型中肿瘤的生长和转移,并可对癌症治疗中癌细胞的变化进行实时观测和评估。活体生物发光成像能够无创伤地定量检测小鼠整体的原位瘤、转移瘤及自发瘤。

2、免疫学与干细胞研究

将荧光素酶标记的造血干细胞移植入脾及骨髓,可用于实时观测活体动物体内干细胞造血过程的早期事件及动力学变化。有研究表明,应用带有生物发光标记基因的小鼠淋巴细胞,检测放射及化学药物治疗的效果,寻找在肿瘤骨髓转移及抗肿瘤免疫治疗中复杂的细胞机制。应用可见光活体成像 原理标记细胞,建立动物模型,可有效的针对同一组动物进行连续的观察,节约动物样品数,同时能更快捷地得到免疫系统中病原的转移途径及抗性蛋白表达的改变。

3、细胞凋亡

当荧光素酶与抑制多肽以融合蛋白形式在哺乳动物细胞中表达,产生的融合蛋白无荧光素酶活性,细胞不能发光,而当细胞发生凋亡时,活化的caspase-3在特异识别位点切割去掉抑制蛋白,恢复荧光素酶活性,产生发光现象,由此可用于观察活体动物体内的细胞凋亡相关事件。

4、病毒侵染

以荧光素酶基因标记的HSV-1病毒为例,可观察到HSV-1病毒对肝脏、肺、脾及淋巴结的侵和病毒从血液系统进入神经系统的过程。多种病毒,腺病毒,腺相关病毒,慢病毒,乙肝病毒等,已被荧光素酶标记,用于观察病毒对机体的侵染过程。

5、基因治疗

基因治疗包括在体内将一个或多个感兴趣的基因及其产物安全而有效的传递到靶细胞。可应用荧光素酶基因作为报告基因用于载体的构建,观察目的基因是否能够在试验动物体内持续高效和组织特异性表达。这种非侵入方式具有容易准备、低毒性及轻微免疫反应的优点。荧光素酶基因也可以插入脂质体包裹的DNA分子中, 用来观察脂质体为载体的DNA运输和基因治疗情况。

6、药物开发

利用标记好的细菌在动物体内对药物的反应,医药公司和研究机构可用这种成像技术进行药物筛选和临床前动物实验研究。

7、组织特异性基因表达

将目标荧光素酶基因或荧光蛋白基因插入到目标染色体DNA上,实现组织特异性的表达。

8、基因工程

为研究目的基因是在何时、何种刺激下表达,将荧光素酶基因插入目的基因启动子的下游,并稳定整合于实验动物染色体中,形成转基因动物模型。利用其表达产生的荧光素酶与底物作用产生生物发光,反应目的基因的表达情况,从而实现对目的基因的研究。可用于研究动物发育过程中特定基因的时空表达情况,观察药物诱导特定基因表达,以及其它生物学事件引起的相应基因表达或关闭。

9、各种疾病模型

研究者根据研究目的,将靶基因、靶细胞、病毒及细菌进行荧光素酶标记,同时转入动物体内形成所需的疾病模型,包括肿瘤、免疫系统疾病、感染疾病等等。可提供靶基因在体内的实时表达和对候选药物的准确反应,还可以用来评估候选药物和其它化合物的毒性。为药物在疾病中的作用机制及效用提供研究方法。

独家4D Real-Time 实时成像技术。传统的小动物活体光学成像,将待测小动物放入成像舱后,等待曝光时间后拍照,获得一张图片作为最终结果。而Biospace Lab PhotonIMAGER 小动物活体光学系统独家的4D Real-Time 实时成像技术,从小动物进入成像舱开始就进行实时拍摄,做到第一时间采集,避免丢失任何光学信息,并做到全局可视化,随时观察信号的积累和整体的变化过程。最终获得一段视频作为结果,保留了信号的动力学数据。传统的成像方式中,成像时间通常依据标准曲线来决定,然而标准曲线无法考虑到不同小动物之间的个体差异,而Biospace Lab PhotonIMAGER 可以实时的展现信号的高峰和平台期,实时定量捕捉微小的信号变化。独家4D Real-Time 实时成像技术还可以避免图片的过曝,当小动物组间差异过大时,为了呈现出低信号组的图像,往往需要增加曝光时间,从而导致高信号组图片过曝。然而4D Real-Time 实时成像技术避免了这样的问题,实时成像中时间分辨率更高,相邻两次拍摄时间更短,并且在在每一时间节点上快速摄取图像,完整记录信号变化,保留真实信号变化。



2、 独家5D清醒成像技术。4D Real-Time 实时成像功能得益于Biospace Lab的核心技术:iCCD技术。使得Biospace Lab有着比市面上同类产品更高的灵敏度(37 ph/s/sr/cm2)、更快的时间分辨率(66 fps)以及更高的信噪比,也因此,Biospace Lab PhotonIMAGER可以做到5D小动物清醒成像。在使用5D小动物清醒成像模式时,小动物在成像暗舱内自由活动,小鼠无麻醉不保定,做到真正的自然状态,消除麻醉对小鼠大脑、神经、代谢活动的影响,降低小鼠的死亡风险,暗室活动舱也避免了小鼠的应激状态,在神经学、代谢、动物行为学领域提供更广泛的应用。

3、 更真实的3D成像技术。Biospace Lab PhotonIMAGER 3D成像模块一次拍摄可以得到四个正交平面的空间信息,之后激光扫描器扫描小动物表面的拓扑学结构,再通过重构的方法整合为3D图像。与一般的3D图像相比,Biospace Lab PhotonIMAGER 3D更真实,四个面的光学信号都是相机真实捕捉的图像,而非通过计算机模拟。
